Mertens and Gagliardini picked up yellow cards, automatically activating a one-match ban.
It means both will miss out on the upcoming showdown at San Siro.
Atalanta defender Rafael Toloi is suspended against Parma and Brescia midfielder Daniele Dessena skips Lazio.
Milan doctor Stefano Mazzoni was given a one-game touchline ban and fined €5,000 for insulting the referee.
That incident saw him sent off from the bench during Friday’s 1-1 draw with Atalanta.
